Sequencing services on Illumina Sequencers

4 colour sequencer, suitable for sequencing low diversity libraries. Flow cells have 1 lane. Maximum yield on four types of flow cells is 1 million read pairs to 25 million read pairs. Maximum read length is 600 cycles to enable sequencing 2 X 300 bases.

2 colour sequencer, suitable for quality check of libraries prior to sequencing on NextSeq. Flow cells have 1 lane. Two types of flow cells are available mid-output and high output and maximum yield is 8 million read pairs and 25 million read pairs, maximum read length is 300 cycles to enable sequencing 2 X 150 bases Not suitable for low diversity libraries.

2 colour sequencer. Sequences on a 4 lane flow cell where 1 pool of library is sequenced across 4 lanes. Maximum yield on two types of flow cells is 130 million read pairs to 400 million read pairs, maximum read length is 300 cycles Not suitable for low diversity libraries.

1 colour sequencer with a patterned flow cell. Sequences on a 1 lane flow cell. Suitable for small microbial whole genome sequencing, 16S sequencing with V4 primers and quality checking libraries prior to sequencing on NovaSeq6000. Maximum yield on one type of flow cell is 4 million read pairs, maximum read length is 300 cycles.

2 colour sequencer with a patterned flow cell. Four types of flow cells are available with 2 lanes (SP, S1 and S2) or 4 lanes (S4). A single pool of libraries can be sequenced across all 2 or 4 lanes of a flow cell using the standard workflow. Alternately, libraries can be split by lane such that a single pool of libraries is sequenced on 1 lane using the XP workflow. Maximum yield on four types of flow cells is 800 million to 10,000 million read pairs. Maximum read length is 300 cycles. Can sequence read length of 500 cycles on a flow cell with 2 lanes and maximum 800 million reads. Shorter fragments are preferentially sequenced. Creates optical duplicates.

Training is available on operation of instruments at locations as specified below. Once trained, you will have access to a calendar to enable scheduling of instruments for your use. Instrument use incurs a fixed cost

1. Qubit 4 at both TKI and HPIMR labs

This instrument is to measure concentration of extracted nucleic acids and libraries.

2. TapeStation 4200 at both TKI and HPIMR labs

This instrument is to assess quality of extracted nucleic acids via DNA Integrity Number (DIN) for DNA and RNA Integrity Number equivalent (RINe) for RNA and high throughput fragment sizing of libraries. 

3. Bioanalyzer 2100 at TKI lab

This instrument is for assessing quality of extracted RNA via RNA Integrity Number (RIN) and low throughput fragment sizing of libraries.

4. Eppendorf Vacuum concentrators for 96 well plates at HPIMR labs

This instrument is to reduce the volume of samples in 96 well plates. It is available in the pre PCR lab as well as the post PCR lab at Perkins site.

If you plan to submit prepared libraries and need guidance on which Illumina sequencer and flow cell are most appropriate, please send us an email with the following details

  1. Library type (RNA-seq, Amplicon-seq, WES, WGS, ATAC-seq WGBS, ChIP-seq etc)
  2. Number of tubes of pooled libraries you wish to submit
  3. Expected reads per pooled library
  4. Sequencing format required ie 2 X 75 or 2 X 100 etc
